Staphylococcal Protein A (Protein A) is a 42kDa MSCRAMM (Microbial Surface Components Recognizing Adhesive Matrix Molecules) originally identified on the cell wall of Staphylococcus aureus. Owing to its high affinity and specificity for the Fc region of antibodies, Protein A is widely used in the biopharmaceutical industry for large-scale antibody production. In biopharmaceutical production, Protein A used for antibody production is typically immobilized on chromatography resins. However, trace amounts may leach into the eluate, potentially contaminating the final antibody product. Protein A can interact with multiple host components, suggesting a potential role as a virulence factor in Staphylococcus aureus infections. Accordingly, the Chinese Pharmacopoeia requires strict monitoring of residual Protein A. To ensure the quality of antibodies purified by Protein A affinity chromatography, anti–Protein A antibodies and ELISA kits have been developed. In ELISA assays, this antibody binds specifically to Protein A and is suitable for detecting residual Protein A in therapeutic proteins and during antibody purification. |
