Principle of the Assay |
This ELISA kit employs an indirect competitive immunoassay principle. The microplate is pre-coated with kanamycin, and standards (or samples) together with kanamycin monoclonal antibody are added simultaneously. The coated kanamycin antigen competes with kanamycin present in standards (or samples) for binding sites on the monoclonal antibody. After incubation at room temperature, unbound components are washed away. An HRP-conjugated secondary antibody is then added to bind to the captured kanamycin monoclonal antibody. A second incubation and wash step are performed to remove unbound substances. Finally, color development is carried out with TMB substrate, and the sample absorbance of the sample is negatively correlated with residual kanamycin concentration in the sample. The kanamycin concentration in the sample can be calculated by matching the absorbance value against the standard curve and multiplying by the corresponding dilution factor. |
