Principle of the Assay

PAF ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-PAF antibody and an PAF-HRP conjugate. The assay sample and buffer are incubated together with PAF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The result of the enzyme-substrate reaction is a complex that is blue in hue. The process is finally stopped by adding a stop solution, causing the solution to turn yellow. In a microplate reader, the color intensity is measured spectrophotometrically at 450 nm. Due to competition for the anti-PAF antibody binding site between PAF from samples and PAF-HRP conjugate, the intensity of the color is inversely proportional to the concentration of PAF. As more sites are taken up by PAF from the sample, fewer sites are left that can bind PAF-HRP conjugate since the number of sites is restricted. A standard curve is drawn connecting the color's intensity (O.D.) and standard concentration. This standard curve is interpolated to determine the PAF concentration in each sample.
