Principle of the Assay

A competitive enzyme immunoassay approach is used in the HIF 1Alpha ELISA kit along with an anti-HIF 1Alpha antibody and an HIF 1Alpha-HRP conjugate. For one hour, the HIF 1Alpha-HRP conjugate is incubated with the assay sample and buffer in a pre-coated plate. The wells are decanted and washed five times when the incubation period is over. The HRP enzyme substrate is then incubated in the wells. The result of the enzyme-substrate reaction is a complex that is blue in hue. The process is finally stopped by adding a stop solution, causing the solution to turn yellow. In a microplate reader, the color intensity is measured spectrophotometrically at 450 nm. The intensity of the color is inversely proportional to the HIF 1Alpha concentration since HIF 1Alpha from samples and HIF 1Alpha-HRP conjugate compete for the anti-HIF 1Alpha antibody binding site. Since the number of sites is limited, as more sites are occupied by HIF 1Alpha from the sample, fewer sites are left to bind HIF 1Alpha-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HIF 1Alpha concentration in each sample is interpolated from this standard curve.
