Principle of the Assay |
IL-1β ELISA kit applies the solid phase sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with an antibody specific to IL-1β. Standards or samples are then added to the microtiter plate wells and IL-1βif present, will bind to the antibody pre-coatedwells.Then a biotin-conjugateddetection antibody specific to IL-1βis added to the wells to form a pre-coatedantibody-antigen-detection antibody Complex. After washing, Streptavidinconjugated Horseradish Peroxidase (HRP)is added to the wells, and then a TMB substrate solution is added to the wells to incubate. TMB substrate solution turns to blue after oxidating by HRP, and finally turns to yellow after adding the stop solution. Color develops in proportion to the amount of IL-1βbound in theinitial steps. The color is measured spectrophotometrically at a wavelength of 450nm. The concentration of IL-1βin the samples is then determined by comparing the O.D. of the samples to the standard curve. |
