Principle of the Assay |
IFNγ ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-IFNγ antibody and an IFNγ-HRP conjugate. The assay sample and buffer are incubated together with IFNγ-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The result of the enzyme-substrate reaction is a complex that is blue in hue. The process is finally stopped by adding a stop solution, causing the solution to turn yellow. In a microplate reader, the color intensity is measured spectrophotometrically at 450 nm. Due to competition for the anti-IFNγ antibody binding site between IFNγ from samples and IFNγ-HRP conjugate, the intensity of the color is inversely proportional to the concentration of IFNγ. As more sites are taken up by IFNγ from the sample, fewer sites are left that can bind IFNγ-HRP conjugate since the number of sites is restricted. A standard curve is drawn connecting the color's intensity (O.D.) and standard concentration. This standard curve is interpolated to determine the IFNγ concentration in each sample. |
