Principle of the Assay |
cGMP ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-cGMP antibody and an cGMP-HRP conjugate. The assay sample and buffer are incubated together with cGMP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. The process is finally stopped by adding a stop solution, causing the solution to turn yellow. In a microplate reader, the color intensity is measured spectrophotometrically at 450 nm. Due to competition between cGMP from samples and cGMP-HRP conjugate for the anti-cGMP antibody binding site, the intensity of the color is inversely correlated with the concentration of cGMP. As more sites are taken up by cGMP from the sample, fewer sites are left that can bind cGMP-HRP conjugate since the number of sites is restricted. A standard curve is drawn connecting the color's intensity (O.D.) and standard concentration. |
